Location of role Oxford, UK Department Proteomics Key Responsibilities Purpose of the role We have an opportunity in our Research Group for a 12-month placement (Year in industry). We are looking for enthusiastic and highly motivated individuals who would relish the chance to learn from Immunocore’s cutting edge science and corporate growth. Immunocore’s ImmTAX technology targets peptides presented by HLA complexes (pHLA) on the cell surface. Mass spectrometry (MS) remains the only direct method to identify and quantify these pHLA targets. However, the limited availability of clinical sample material poses a significant challenge for MS-based analysis, especially in the context of informing patient selection and understanding the mechanism of target pHLA presentation i.e. protein expression to degradation to cell surface presentation. We aim to deepen our understanding of this process by quantifying both the absolute levels of target pHLA and the proteins involved in the pHLA presentation machinery, as well as the source proteins from which these targets originate in different biological models such as cell lines, patient derived xenografts (PDX), and tumour samples. By integrating these datasets, we aim to establish predictive relationships to support translational decision-making in clinical contexts. Key Responsibilities Receive training/mentorship and develop a set of skills (such as: sample preparation, mass spectrometry, data analysis) that will allow identification and absolute quantification of pHLA, and proteins using mass spectrometry based techniques. Grow relevant cell lines and process complex biological systems such as PDX and tumour samples to conduct immunopeptidomics and proteomics experiments for identification and quantification of HLA peptides, and proteins. Analyse mass spectrometry data using bioinformatics tools developed in house and state of the art software to uncover biology and develop the predictive tools for pHLA presentation. Develop deep understanding of related scientific literature. Education & Qualifications Undertaking (currently in second year and looking for a year in industry) BSc. in either Chemical, Biological or Biomedical Sciences with an analytical chemistry, biochemistry or immunology component.
