Why join us?
Parabilis Medicines is a distinctive biopharmaceutical company aiming to become a leader in therapeutics. Our platform, Helicon therapeutics, involves peptides capable of efficient cell entry and modulation of protein-protein and protein-DNA interactions. This technology has the potential to revolutionize treatment options by targeting intracellular disease drivers previously considered undruggable.
Our lead program, FOG-001, is an early-stage, first-in-class TCF-blocking β-catenin inhibitor, currently in clinical trials for advanced solid tumors, including colorectal cancer. We leverage proprietary machine learning and computational methods in our discovery process. With over $500 million raised, Parabilis is headquartered in Cambridge, MA.
What’s the opportunity?
We seek a skilled biochemist/cell biologist with drug discovery experience to join our DeNovo Discovery team. The role involves developing assays to study cellular uptake and endosomal escape of our therapeutics, collaborating with chemists, biologists, and data scientists to design experiments assessing Helicon uptake, cytosolic exposure, and escape mechanisms.
You will also support efforts to identify new molecules by performing routine testing for cytosolic exposure and cellular uptake.
Key objectives include:
* Identify and refine core assays to measure and optimize cytosolic exposure of Helicons
* Develop additional assays to contextualize CE data
* Provide data-driven insights to inform drug design and structure-activity relationships
* Design cell-based functional assays using techniques like cell fractionation, mass spectrometry, and confocal microscopy
What you'll need to be successful:
* Ph.D. in relevant fields with 3+ years of experience, or Master's with 8+ years in drug discovery
* Experience with liquid chromatography-mass spectrometry
* Experience developing cell-based uptake and proximity-dependent activity assays (e.g., NanoBiT, NanoBRET) is a plus
* Knowledge of biophysical/biochemical assays (ITC, DSF, MST, FP, TR-FRET) and structural biology is advantageous
* Experience in oncology and peptide drug discovery is preferred
* Strong motivation, independence, organization, and multitasking skills
* Flexibility to adapt to changing workflows and projects
* Excellent interpersonal and communication skills
On-site work and in-person meetings are required.
