Applications are invited for a Research Associate position to work with Dr Joan Chang (MRC CDA Research Fellow) at the Division of Molecular and Cellular Function (MCF, ), and Manchester Cell-Matrix Centre (MCMC, ). This is an exciting project aiming to elucidate the molecular mechanisms to collagen trafficking and regulation, and how fibroblasts and myeloid cells coordinate to form this matrix, in the context of healthy and fibrotic tissues. Collagen is one of the most abundant proteins in the body, and dysregulation leads to a wide range of pathologies such as fibrosis and cancer metastasis. Understanding how cells regulate collagen will allow better understanding of collagen pathologies. We have recently identified that 1) collagen deposition is under circadian control, 2) collagen protomeric secretion and fibrillogenesis are separate pathways, 3) endocytic recycling of external collagen is central to fibrillogenesis, and 4) myeloid cells also produce a collagenous matrix.

Here we will further map out the molecular networks involved in controlling the switch between monomeric secretion and fibril formation, and characterise an endogenously-tagged collagen mouse model developed within the lab. The project will employ a range of techniques, including mass spectrometry and flow cytometry analyses, state-of-the-art imaging techniques (e.g. super-resolution, confocal, electron microscopy), and cells expressing tagged proteins to obtain new mechanistic insights. These findings will be clinically valuable in multiple collagen-diseases, including lung fibrosis and chronic skin wounds.

You will interact closely with the principal investigator and will be expected to contribute intellectually and take ownership of the project. In addition, although you will be based in Manchester, short visits to collaborators may be required for data acquisition and project development. As previously we have identified circadian rhythm as a key regulator, you may need to perform circadian time courses which meant overnight sample collection. Finally, you will also be expected to attend appropriate training and workshops in order to strengthen and expand your skill sets.

You will have a Ph.D. degree or equivalent in cell biology, molecular biology, biochemistry or a related area of research. You will have experience in using Mass Spectrometry to study proteome changes. Experience of biochemical techniques such as protein expression/purification, SDS-PAGE and Western Blotting, flow cytometry and microscopy are also desirable. Knowledge of extracellular matrix would be an advantage.

Work will be based at the MCF, and will be in close collaboration with the Core Facilities, and MCMC. Duties will include design of experiments, data analysis, presentation of work and preparation of papers for publication, as well as some supervision of students.

This is a full-time fixed term appointment starting 13 October 2025, or as soon as possible thereafter, and is funded until 31 March 2027.
